Description

What is MT2?

MT2 is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2Structurally, the compound belongs to the N-terminally acetylated, C-terminally amidated peptides and is known by CAS number 121062-08-6 with the molecular formula C50H69N15O9 (approximately 1024 Da, PubChem CID 92432).

Two chemical interventions determine the character of the molecule within its peptide class. The first is the lactam bridge between the side chains of the aspartic acid and lysine residues: this closes the chain into a ring and fixes the backbone in a defined conformation. The second is the incorporation of non-natural residues—norleucine and the D-enantiomer of phenylalanine—which make the chain significantly less susceptible to enzymatic degradation under laboratory conditions. Together, cyclization and D-substitution make MT2 a highly characterizable model compound in comparative peptide research and in analytical studies of conformationally restricted cyclic peptides.

Applications in a laboratory context

Within peptide chemistry and analytical work, MT2 occurs in the following in vitro research areas, among others:

  • Structure-activity relationship research to the contribution of the lactam bridge, N-terminal acetylation and C-terminal amidation to the spatial planning of the ring.
  • Peptide chemistry research regarding solubility, ring strain, and aggregation behavior of cyclic chains, and regarding the influence of the D-configured residue on the conformation.
  • Enzymatic stability studies in which the degradation rate of the cyclized chain in vitro is compared with that of linear analogues with the same residue composition.
  • Analytical method development and validation (HPLC, LC-MS, peptide mapping) where the material serves as a reference standard for the separation of cyclic and linear variants and for degradation studies of cyclic peptides.

Shape, handling and storage

The material is supplied as freeze-dried powder (lyophilisate) in a sealed glass vial with a rubber septum and aluminium shrink cap. The lyophilisate is white to off-white and hygroscopic; variations in the shape of the freeze-dried cake are inherent to the freeze-drying process and do not indicate anything about the quality of the material. The vial remains closed and in the original packaging until the material is processed in the laboratory.

Store the unopened lyophilisate at 2–8 °C, in a dark and dry place, protected from light and moisture. Allow a refrigerated vial to reach room temperature before opening to prevent condensation on the powder, and handle the material with the usual laboratory precautions: a clean working environment, gloves and eye protection, and disposal of residual material in accordance with applicable guidelines.

Analytical quality

For a cyclized peptide, documented starting purity is particularly relevant: linear by-products of the ring closure have virtually the same mass as the target compound and are chromatographically close to each other, making them easily confused in a measurement series. Our MT2 is supplied with a purity of ≥ 99%, determined by HPLC analysis in accordance with the accompanying Certificate of Analysis. The available Certificates of Analysis can be viewed on our website. lab results page.
