Description
What is Mazdutide?
Mazdutide is a synthetic acylated polypeptide: a long amino acid chain to which a long-chain fatty acid is covalently linked via a linker on the side chain of a lysine residue. The compound is known under CAS number 2259884-03-0, with the molecular formula C210H322N46O67 and a molecular weight of approximately 4563 Da. This places the molecule within the lipopeptides: peptide chains that carry a covalently bonded fatty acid tail.
This acylation is the defining structural feature and drastically alters the physicochemical profile of the chain. A lipophilic tail attached to an otherwise predominantly polar chain shifts solubility in aqueous media, increases the tendency towards self-association and aggregation with increasing concentration, and increases non-covalent protein binding in vitro. Because the coupling site on the lysine side chain is chemically defined, the compound can be sharply characterized; this makes Mazdutide a useful model compound in comparative peptide research and in analytical investigations into the behavior of long acylated chains.
Applications in a laboratory context
Within peptide chemistry and analytical work, Mazdutide is found in the following in vitro research areas, among others:
- Structure-activity relationship research to the contribution of individual sequence positions and of the acylation site to spatial planning and the rigidity of the chain.
- Peptide chemistry research regarding solubility, amphiphilic behavior and aggregation of acylated chains in aqueous and mixed media, and the influence of pH and ionic strength thereon.
- Enzymatic stability studies in which the degradation rate of the chain in vitro is compared with that of non-acylated analogues to quantify the contribution of the fatty acid tail to chain stability.
- Analytical method development and validation (HPLC, LC-MS, peptide mapping) where the material serves as a reference standard for retention behavior, charge distribution, and fragment patterns of long acylated peptides.

Shape, handling and storage
The material is supplied as freeze-dried powder (lyophilisate) in a sealed glass vial with a rubber septum and aluminium shrink cap. The freeze-dried cake is white to off-white and may vary in density per vial; this variation is inherent to the freeze-drying process and says nothing about the quality of the material. Lyophilisates of acylated peptides are hygroscopic, which is why the vial remains closed and in the original packaging until the material is processed in the laboratory.
Store the sealed lyophilisate at 2–8 °C, in a dark and dry place, protected from light, moisture, and temperature fluctuations. Allow a refrigerated vial to reach room temperature before opening to prevent condensation from forming on the powder. Furthermore, handle the material with the usual laboratory precautions: work in a clean environment, wear gloves and eye protection, and dispose of residual material according to applicable guidelines.
Analytical quality
In the case of a long acylated chain, the identity and purity of the starting material determine how interpretable a measurement series is: deletion sequences and non-acylated variants closely resemble the target compound yet yield an anomalous analytical signal. Our Mazdutide is supplied with a purity of ≥ 99%, determined by HPLC analysis in accordance with the accompanying certificate of analysis. The available certificates of analysis can be viewed on our website. lab results page.
