What is Gonadorelin?

Gonadorelin is the synthetic form of gonadotropin-releasing hormone (GnRH, also referred to as LHRH) and consists of ten amino acids in the sequence pGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH₂The two ends are chemically unique. At the N-terminus, there is no free amine but a pyroglutamic acid: an intramolecularly cyclized glutamine in which the amino group is incorporated into a lactam ring. At the C-terminus, there is no free carboxylate but a glycinamide. Both ends are therefore electrically neutralized, meaning the net charge is determined entirely by the side chains, in practice primarily by the arginine at position 8 and the histidine at position 2. This charge neutralization simultaneously reduces the binding sites for amino- and carboxypeptidases.

In research models, Gonadorelin is used as a reference ligand for the GnRH receptor, a class A G-protein-coupled receptor. Because this peptide is the best-characterized ligand for that receptor, it serves as a reference point in binding studies using pituitary cell culture systems and membrane preparations when comparing modified analogs. Structural research focuses on the conformation in the binding groove: the glycine at position 6 allows for a tight bend.
