Descrizione

VIP (vasoactive intestinal peptide) is a naturally occurring neuropeptide, chemically a linear polypeptide of 28 amino acids belonging to the secretin-glucagon peptide family. In scientific research, it is studied as a model compound for signaling via class BG protein-coupled receptors and as a reference standard in analytical work. This compound is offered as a research kit containing 10 mg of freeze-dried VIP, exclusively for laboratory and research purposes.

Che cos'è un VIP?

VIP is a linear peptide with the sequence His-Ser-Asp-Ala-Val-Phe-Thr-Asp-Asn-Tyr-Thr-Arg-Leu-Arg-Lys-Gln-Met-Ala-Val-Lys-Lys-Tyr-Leu-Asn-Ser-Ile-Leu-Asn-NH2, in which the C-terminus is terminated as an amide. The molecule belongs to the secretin-glucagon superfamily and is therefore structurally closely related to PACAP and secretin. Characteristic features include its strongly basic nature — the sequence contains multiple arginine and lysine residues — and the tendency towards alpha-helix formation in membrane mimetic environments. This combination of ordered helical structure and well-characterized receptor interaction makes the compound a widely used model in research into peptide receptor recognition.

In research models, VIP binds with nanomolar affinity to VPAC1 and VPAC2, two class B1 G-protein-coupled receptors from the same structural family as the secretin receptor; the affinity for the related PAC1 receptor is significantly lower in vitro. Both VPAC receptors couple to stimulating G-proteins (Gs), after which activation of adenylate cyclase and an increase in intracellular cyclic AMP are measured in cell models, followed by activation of protein kinase A. This highly reproducible cAMP response makes the peptide useful as a positive control in receptor pharmacological test systems. Consequently, VIP is used as a model compound and reference substance in scientific research.

Aree di ricerca

In a laboratory context, VIP occurs in the following research areas, among others:

  • Receptor research into VPAC1 and VPAC2 binding and receptor selectivity within the PACAP/VIP receptor family, in membrane preparations in vitro.
  • Ricerca su Gs-coupled signal transduction, adenylate cyclase activity and cAMP formation in cell culture models.
  • Enzymological research into the degradation of the peptide by peptidases such as dipeptidylpeptidase-4 and neprilysin.
  • Biophysical modeling studies of alpha-helix formation and conformation in membrane mimetic systems (CD spectroscopy, NMR).
  • Analytical method development and validation (HPLC, LC-MS) where VIP serves as the reference standard.
  • Studi di stabilità e solubilità di peptidi liofilizzati in condizioni controllate.

Ciascuna di queste applicazioni prevede ricerche controllate in laboratorio o su modelli preclinici.

VIP within our research range

VIP falls under the category of research kits for nasal administration models within our range, in which a freeze-dried peptide is combined with a sterile physiological saline solution and a spray bottle. Researchers working with this category regularly compare the compound with other peptides studied via their own biochemical target. While VIP focuses on the VPAC receptors and the associated cAMP pathway, other components within the same product line have been chosen specifically to cover divergent receptor families or enzymatic targets. These kits share the same design and presentation form but differ fundamentally in molecular class, peptide length, and investigated mechanism.

Forma, manipolazione e stoccaggio

VIP is supplied as a freeze-dried powder in a sealed vial containing a standardized quantity of 10 mg. The kit also contains a sterile physiological saline solution (0.9% NaCl) and a spray bottle. Unlike solid tablet forms, dissolution is required for a lyophilisate before the material can be processed in an analytical or cell culture setup.

Store the freeze-dried powder in a cool, dry place, protected from direct light, in the original packaging; once dissolved, peptides are more sensitive to temperature changes and should be stored in a cool, dark place. Handle the compound with standard laboratory precautions: work in a clean environment, use appropriate personal protective equipment, and prevent cross-contamination with other research materials. Keep the packaging out of reach of unauthorized persons.

Composizione e risultati di laboratorio

The lyophilisate in this kit has a controlled purity of ≥ 99% (HPLC). The supplied liquid is a sterile physiological saline solution (NaCl 0.9%) and contains no active compound itself.

Domande frequenti

To which fabric class does VIP belong?
VIP is a 28-amino acid neuropeptide of the secretin-glucagon peptide family. It is therefore structurally related to PACAP and secretin and is not a small synthetic molecule.

Which receptors are studied in research with VIP?
In research models, the peptide binds with nanomolar affinity to the class B1 receptors VPAC1 and VPAC2. Both couple to Gs-proteins, after which adenylate cyclase activity and cAMP formation are measured in cell models; the affinity for the PAC1 receptor is significantly lower in vitro.

In quale forma viene fornito il composto?
As a freeze-dried powder of 10 mg in a sealed vial, together with a spray bottle and a sterile physiological saline solution (NaCl 0.9%).

How do I store the kit?
The freeze-dried powder should be stored in a cool, dry, and dark place in the original packaging; material that has been dissolved should be kept cool and protected from light.
