Description
What is AOD-9604?
AOD-9604 is a synthetic linear peptide of sixteen amino acid residues with the sequence Tyr-Leu-Arg-Ile-Val-Gln-Cys-Arg-Ser-Val-Glu-Gly-Ser-Cys-Gly-Phe. The chain consists of the fifteen residues of a defined C-terminal fragment of a longer, naturally occurring polypeptide chain, supplemented by one additional tyrosine at the N-terminal position. The compound is known under CAS number 221231-10-3, with the molecular formula C78H123N23O23S2 (approximately 1815.1 g/mol) and PubChem CID 71300630.
The spatial structure is determined by the two cysteine residues at positions 7 and 15, which form a disulfide bridge with each other. This bridge closes part of the sequence into a loop, giving the chain in solution a significantly more limited conformation range than a fully open chain of comparable length. The combination of a short, fully written-out sequence and exactly one internal bridge makes AOD-9604 a highly reproducible model compound in comparative peptide research: the closed and reduced forms differ by two mass units and can be separated chromatographically, making the substance useful in research into the formation, opening, and reoxidation of disulfide bridges.
Applications in a laboratory context
Within peptide chemistry and analytical work, AOD-9604 occurs in the following in vitro research areas, among others:
- Structure-activity relationship research to the contribution of the added N-terminal tyrosine and of both cysteine positions to chain conformation, charge distribution, and chromatographic behavior.
- Peptide chemistry research towards solubility and aggregation tendency of short chains closed by a disulfide bridge in aqueous and mixed media.
- Enzymatic stability studies in which the breakdown of the chain is monitored under defined pH, temperature, and enzyme conditions, and the formed fragments are characterized by mass spectrometry.
- Analytical method development and validation (HPLC, LC-MS, peptide mapping) where the tyrosine residue provides a usable UV signal and the closed form serves as a reference peak alongside the reduced variant.

Shape, handling and storage
The material is supplied as freeze-dried powder (lyophilisate), white to off-white in color, in a sealed glass vial with a rubber septum and aluminum shrink cap. Short peptide lyophilisate is hygroscopic; therefore, the vial remains closed in the original packaging until the sample is taken into processing in the laboratory.
Store the sealed vial freeze-dried at 2–8 °C, in a dark and dry place. The usual laboratory precautions apply: work in a clean, dry environment, avoid repeated temperature changes, and record the batch number and opening date for each vial in the lab logbook so that measurement series remain traceable afterwards. For chains containing a disulfide bridge, contact with reducing agents requires extra attention, as this alters the conformational profile of the sample.
Analytical quality
In comparative peptide analysis, the purity of the starting material determines how reliably the measurement results can be interpreted: shortened sequences and degradation products generate additional chromatographic peaks that may coincide with the main component. Our AOD-9604 is supplied with a purity of ≥ 99%, determined by HPLC analysis in accordance with the accompanying Certificate of Analysis. The available Certificates of Analysis can be viewed on our website. lab results page.
